Morgan Stanley recently released a research report covering eight biotechnology companies for the first time, whose businesses involve rare diseases, ophthalmology, pulmonary diseases, and neuropsychiatric disorders.

According to Wall Street Insights APP, Morgan Stanley recently released a research report initiating coverage on eight biotech companies involved in rare diseases, ophthalmology, pulmonary diseases, and neuropsychiatric conditions. The firm is particularly optimistic about the growth potential of PTC Therapeutics (PTCT.US), driven by Sephience, as well as the opportunity for Belite Bio (BLTE.US) to become the first approved and marketed therapy for Stargardt disease. Meanwhile, attention is given to Trevi Therapeutics (TRVI.US) for its market prospects in chronic cough and to COMPASS Pathways (CMPS.US) for its potential commercialization phase beginning in 2027.

PTC Therapeutics—‘Overweight’ rating, Target Price $104

Morgan Stanley believes that the oral phenylketonuria (PKU) drug Sephience, developed by PTC Therapeutics, surpassed $600 million in annualized sales in just one year since launch, with future peak revenue expected to reach around $1.7 billion. Combined with approximately $2.2 billion in cash and equivalents as of June 30, 2026, the stock has upside potential even if no value is assigned to its R&D pipeline.

Currently, about 100 initial U.S. centers of excellence have all begun prescribing Sephience. Morgan Stanley notes that the most contested market issues are the rate of new net patient additions and the continuity of patient treatment. For the Q3 results released on October 29, if the net new patient additions are on par with Q2’s 403 patients and the company raises its FY2026 product revenue guidance, this would represent a baseline result. The bank expects Sephience FY2026 revenue to reach $955 million, above the company’s current guidance range of $850-$950 million.

Morgan Stanley assigns only a moderate valuation to the R&D pipeline. Beyond the marketed products, potential catalysts include: engaging with the FDA on votoplam in H2 2026, initiation of the PROVE-FA study, and anticipated completion of the ST-920 Biologics License Application (BLA) submission by Q4 2026.

Belite Bio—‘Overweight’ rating, Target Price $206

Morgan Stanley believes that tinlarebant could become the first approved therapy for type 1 Stargardt disease (STGD1), with a PDUFA goal date of February 12, 2027. This is based on Phase III DRAGON trial data showing a 35.7% slower lesion growth rate (p=0.0033) with a favorable safety profile, consistent with its mechanism.

Although approval may rely on just one Phase III trial, the bank believes that additional evidence from the DRAGON trial—including fellow-eye data, DAF, and qAF measures—provides further support. Management’s description of FDA feedback remains constructive. The bank anticipates that the FDA will not require DRAGON II trial data.

Morgan Stanley sees multiple blockbuster-level market opportunities in the STGD1 space, with risk-adjusted peak revenue around $3 billion. About 20,000 patients in the United States have a clinical diagnosis. Final pricing relative to the bank’s $300,000 wholesale acquisition cost (WAC) assumption could also lend further upside.

In H1 2027, interim data from the PHOENIX study for geographic atrophy (GA) will be released. The bank gives this project a 40% probability of success (PoS), which could provide upside beyond the STGD1 business.

Trevi Therapeutics—‘Overweight’ rating, Target Price $23

Morgan Stanley believes the dual μ-opioid receptor agonist/κ-opioid receptor antagonist mechanism of Haduvio positions Trevi Therapeutics to become the first FDA-approved provider for chronic cough therapy. The firm believes there is a relatively low-risk blockbuster-level opportunity in idiopathic pulmonary fibrosis (IPF)-related cough, which does not depend on the refractory chronic cough (RCC) business. The latter’s Phase IIb LAKE trial may further unlock multiple blockbuster-level market potentials.

Morgan Stanley expects that in the Phase III OCEAN-2 trial (data expected in H2 2027), if 24-hour cough frequency is reduced by at least about 30% versus placebo, with supporting patient-reported outcomes (PROs) and a discontinuation rate similar to placebo, this would meet baseline expectations. This is aligned with statistical design assumptions and the Phase III CORAL study’s observation of a 36.5% decrease in the 54 mg BID group.

Before the data readout, key milestones include a reassessment of LAKE trial sample size for RCC indication (expected Q4 2026) and agreement with the FDA on the non-IPF interstitial lung disease (ILD) indication (expected in H2 2026). The OCEAN-1 trial is expected to report data in H1 2028, providing an assessment of sustained efficacy over 24 weeks.

Compass Pathways—‘Overweight’ rating, Target Price $18

Morgan Stanley believes COMP360 could become the first approved classic psychedelic therapy. The company’s rolling NDA submission for treatment-resistant depression (TRD) is anticipated to complete in Q4 2026, with a competitive National Priority Review Voucher (CNPV) supporting potential approval in H1 2027.

The filing is based on two positive Phase III studies: COMP005 showed a single dose yielded a 3.6-point lower Montgomery–Åsberg Depression Rating Scale (MADRS) score versus placebo; COMP006 showed a 3.8-point difference after two doses relative to 1 mg active control; both studies achieved p-values <0.001. The therapy is well-tolerated with no imbalances in clinically concerning suicidal tendencies.

Given the need for approximately six hours of supervised treatment and the requirement for DEA re-scheduling for commercialization, launch execution remains a key debate. However, Morgan Stanley believes the network of roughly 8,000 Spravato treatment centers, dedicated psychedelic CPT codes, and management’s prudent “go slow to go fast” strategy are supportive of commercialization. The firm expects revenue to gradually ramp up in H2 2027 rather than spike at launch.

Morgan Stanley sees a blockbuster-level market opportunity in TRD, with risk-adjusted peak U.S. sales of about $1.3 billion. There are around four million TRD patients in the U.S., while Spravato currently treats only about 100,000. Management anticipates 2-4 doses per patient per year, higher than the firm’s 1.75-dose assumption, adding potential upside.

The Phase IIb/III COMP202 study for post-traumatic stress disorder (PTSD) is not included in the base case valuation but could bring further upside beyond the TRD business.

4D Molecular Therapeutics (FDMT.US)—‘Equal Weight’ rating, Base Case Target Price Range $7–$16

Morgan Stanley notes that the single intravitreal injection of 4D-150 could substantially reduce the injection burden of anti-VEGF therapies. Frequent injections are a key reason for real-world underperformance of such treatments, and 4D-150 may improve outcomes without requiring superior efficacy. However, beyond first-year durability and the 20-week steroid taper regimen are critical to broader adoption.

Morgan Stanley believes that in the Phase III 4FRONT-1 trial (data expected Q2 2027), if best-corrected visual acuity (BCVA) is non-inferior to 2 mg aflibercept every 8 weeks (Q8W) within a non-inferiority margin of about four letters, intraocular inflammation (IOI) rate matches the 3% seen in the PRISM study (mild, transient), and treatment burden is reduced by over 70-80%, this would meet expectations.

Beyond trial data, the bank will closely watch the outcome of the AbbVie/Regenxbio subretinal suravec Phase III trial (data expected Q4 2026) and Lilly’s Ixo-vec ARTEMIS trial (data expected Q1 2027).

Avalyn Pharma (AVLN.US)—‘Overweight’ rating, Target Price $57

Morgan Stanley believes that inhaled drug administration could resolve previous tolerability issues with Esbriet and Ofev. For AP01 and AP02, the firm sees potential for both drugs to achieve multiple blockbuster status without relying on efficacy superior to existing treatments.

Morgan Stanley expects that in the Phase IIb MIST study (data expected H2 2027), if forced vital capacity (FVC) shows a significant difference versus placebo—at least about 65 mL—and discontinuation rates are similar to placebo, the results would meet baseline expectations.

Beyond trial data, key events include the Bristol Myers Squibb ALOFTPPF trial (data expected early 2027) and exercise of the PARI option (expected by January 1, 2028).

Regenxbio (RGNX.US)—‘Overweight’ rating, Target Price $18

Morgan Stanley believes Regenxbio has two late-stage, single-administration gene therapies driving value: suravec, developed in collaboration with AbbVie for wet age-related macular degeneration (wet AMD) and diabetic retinopathy (DR); and wholly-owned RGX-202 for Duchenne muscular dystrophy (DMD).

The most critical near-term milestone is the release of pivotal trial data for wet AMD. Meanwhile, with narrowing Elevidys label scope, the DMD field offers room for a second gene therapy. (SRPT is covered by Mike Ulz.)

Morgan Stanley believes that if the ATMOSPHERE and ASCENT trials (data expected Q4 2026) both meet non-inferiority on BCVA, reduce treatment burden by at least 50%, and show no drug-related IOI, this would count as a positive outcome.

The rolling BLA filing for RGX-202 is expected to be completed in Q1 2027, and the FDA’s review designation is also a key event. Both will impact the chances of accelerated approval in H2 2027.

Key risks include: procedure- or inflammation-related safety signals in Phase III involving more than 1,200 patients; FDA requiring randomized controlled trial data for DMD; and financing risks. The company expects current cash to support operations through Q4 2027 in preparation for potential commercialization.

Kalaris Therapeutics (KLRS.US)—‘Overweight’ rating, Target Price $14

Morgan Stanley believes the heparin-binding design of TH103 could deliver longer durability than current anti-VEGF therapies. Given that its current share price barely reflects TH103’s value, the bank sees an attractive risk/reward profile ahead of the preliminary data from the Ib/II MAD study, expected in H1 2027.

Safety is the main area of debate. Morgan Stanley’s base scenario is that the latest TH103 batch only causes transient, mild to moderate IOI; vision and anatomical improvement is comparable to approved anti-VEGF drugs; and a significant portion of patients reach a retreatment interval of at least 16 weeks.

Beyond trial data, Morgan Stanley will also focus on the AbbVie/REGENXBIO suravec Phase III trial (data expected Q4 2026), and the 4D-150 Phase III 4FRONT-1 trial (data expected Q2 2027), both serving as key benchmarks for durability.

Given that the company expects current cash to support operations only until Q4 2027 and the Phase III trial is planned to start at the end of 2027, the company will likely need to raise additional funds before trial initiation.

The most direct signal that US stocks have peaked: South Korean chip exports?

Currently, the U.S. stock market is supported solely by strong earnings expectations. If semiconductor exports from South Korea and Taiwan decline, it may provide an early warning 3-6 months in advance of capital expenditure peaking. Once expectations reverse, high yields will instantly crush valuations.
